[content title="Description"]Transposable elements (TEs)-DNA sequences that are capable of moving from one chromosome location to another-are found in all living organisms. They have been increasingly investigated in a wide spectrum of species, including bacteria, plants, fungi, and animals. In Mobile Genetic Elements: Protocols and Genomic Applications, leading experts describe in step-by-step detail their most productive transposon-based methods and strategies for studying genome structure, function, and evolution. These readily reproducible techniques cover a broad range, including mutagenesis, transgenesis, gene silencing, and molecular systematics. Among the highlights are a series of DNA hybridization methods for analyzing the distribution and dynamics of mobile DNA at the hosts' genomic level, techniques for studying LTR retrotransposons in heterologous host systems, and mutagenesis protocols for investigating gene functions in a broad range of organisms. Additional methods deal with highly informative sets of polymorphic markers, RNAi technology in gene silencing, and applications during transgenesis. The protocols presented follow the successful Methods in Molecular Biology™ series format, each one offering step-by-step laboratory instructions, an introduction outlining the principle behind the technique, lists of equipment and reagents, and tips on troubleshooting and avoiding known pitfalls.

[content title="Description"]A collection of key cytogenetic and FISH techniques used by modern clinical laboratories in the genetic analysis of human malignancies. The book's practical advice and methods are suitable for use at every level of expertise, including fully established laboratories, but with a sympathetic bias towards anyone considering setting up a new cytogenetics service. Here the reader will find not only elementary tutorials on the fundamentals of human karyotypes and chromosome analysis, but also detailed discussions on how laboratories may optimally upgrade their repertoire of capabilities to include such newer complementary techniques as CGH, FISH, and M-FISH.

[content title="Description"]Psychiatric Genetics provides the reader with a complete view of the methodological problems encountered in psychiatry genetics and proposes solutions to commonly occurring questions. The best European and American specialists have given a thorough review on the advantages and disadvantages of genetic epidemiological methods, the way to choose a genetic marker or a clinical interview and how to ascertain patients, unaffected relatives and controls and what should be the criteria to include a case or a control. New phenotypic methods are described focusing on candidate symptom and endophenotype approaches. Examples coming from cognitive neurosciences, biochemistry, electrophysiology and brain imaging techniques are reviewed. [content title="Description"]Mutations within mitochondrial DNA (mtDNA) and the nuclear genes involved in the maintenance of mitochondrial DNA have been linked to a wide range of human diseases, including several of the most common diseases of aging. In Mitochondrial DNA: Methods and Protocols internationally recognized authorities describe in great detail the methods they have perfected to analyze mtDNA and the proteins involved in its maintenance. The analytical techniques cover the purification of mtDNA from a variety of sources and the analysis of DNA for both deletions, point mutations, and damage, for replication intermediates, and for following the fate of mtDNA outside of the mitochondria. Additional analytical methods are presented for analyzing the proteins and enzymes that maintain mtDNA. Each readily reproducible protocol includes step-by-step instructions, tips on avoiding pitfalls and extending the method to other situation, and introductory material explaining the theory behind the process.
Comprehensive and timely, Mitochondrial DNA: Methods and Protocols offers both basic and clinical researchers proven cutting-edge methods for analyzing the role mtDNA plays in the aging process, apoptosis, and possibly some cancers, and for investigating the cause of mitochondrial dysfunction and disease. [content title="Description"]Stem cell research continues to make headlines with each new biological breakthrough and court ruling, and this entry in the Point/Counterpoint series takes on this crucial contemporary public-policy issue. Science, religion, and ethics are part of the intense debate. Is it immoral to use cells from embryos for scientific research? What if it saves lives? At the center is the issue of when life begins. The text is dense, but the science is accurate, and the ethical arguments give voice to all sides. [content title="Description"]This volume provides an overview of exacerbation models of asthma and chronic obstructive pulmonary disease (COPD). Within this wide field the book focuses on experimental systems that mimic pathobiological processes likely to be critical in exacerbations of these conditions. To generate insight into the mechanisms of exacerbation of pulmonary disease and to promote the discovery of future treatments, both animal models and human experimental models are described. For this update some of the most eminent scientists within the area of pulmonology could be recruited to share their knowledge of this evolving field of human medicine. Models of Exacerbations in Asthma and COPD will be of great interest to pulmonologists, allergologists, specialists in internal medicine and critical care, as well as to microbiologists, infectiologists and pharmacologists studying the response to respiratory infections.

[content title="Description"]Why do racial and ethnic controversies become attached, as they often do, to discussions of modern genetics? How do theories about genetic difference become entangled with political debates about cultural and group differences in America? Such issues are a conspicuous part of the histories of three hereditary diseases: Tay-Sachs, commonly identified with Jewish Americans; cystic fibrosis, often labeled a ''Caucasian'' disease; and sickle cell disease, widely associated with African Americans.

In this captivating account, historians Keith Wailoo and Stephen Pemberton reveal how these diseases -- fraught with ethnic and racial meanings for many Americans -- became objects of biological fascination and crucibles of social debate. Peering behind the headlines of breakthrough treatments and coming cures, they tell a complex story: about different kinds of suffering and faith, about unequal access to the promises and perils of modern medicine, and about how Americans consume innovation and how they come to believe in, or resist, the notion of imminent medical breakthroughs.

With Tay-Sachs, cystic fibrosis, and sickle cell disease as a powerful backdrop, the authors provide a glimpse into a diverse America where racial ideologies, cultural politics, and conflicting beliefs about the power of genetics shape disparate health care expectations and experiences. [/content]

[content title="Description"]The conceptual basis for molecular mimicry was first defined in the early 1980s when monoclonal antibodies against viruses were also shown to react with non-viral host protein; in this case, measles virus phosphoprotein cross-reacted with host cell cytokeratin, herpes simplex virus type 1 with host-cell vimentin and vaccinia virus with host-cell intermediate filaments. Following this discovery, others emerged, again at the clonal level, that T cell clones against proteins from a variety of infectious agents also reacted with host antigenic determinants. The clonal distinction was imperative for the initial definition of mimicry. At least 30 years prior to our initial description of molecular mimicry involving cross-reactions between numerous microbes, on the polyclonal antibody level, streptococcus was believed to react with renal glomeruli, heart and basal ganglia to account for the glomerulonephritis, heart and valvular disease and chorea, respectively. However, subsequent research showed that the nephritis was caused by immune complex deposits and the tissue damage they produced. Later, in 1990, the cross-reactivity of streptococcal antigen with myocardial antigens on a clonal level was uncovered. Hence, for both historical reasons and mechanistic understanding, it is best to provide evidence for cross-reactivity at the clonal level to prove that molecular mimicry exists.
